Topic Structural studies of bacterial transcription regulators

Abstract

Bacterial repressors function as molecular switches: binding of an effector molecule modulates their ability to interact with DNA operator and the gene transcription is turned on or off. Most of transcriptional repressors involved in regulation of metabolic genes in B. subtilis belong to one of the two large families: GntR and DeoR, respectively. Understanding the structure-function relationship and molecular mechanisms by which individual repressors execute their regulatory functions greatly benefits from knowledge of their 3D structures. In this project, we selected several functionally characterized repressors as targets for structural characterization. The crystallographic studies will be combined with complementary biophysical techniques to further characterize effector binding as well as DNA operator recognition.
