Topic Studies of GPCR/G-protein signaling by advanced optical microscopy techniques

Abstract

G-protein coupled receptors and heterotrimeric G-proteins play an important role in cell signaling. G-protein signaling is crucial for cardiovascular function, neurotransmission and many other processes. G-protein signaling is therefore a target of many pharmaceutical drugs, and knowledge of molecular details of G-protein signaling is important both for our basic understanding of cellular processes and for various pharmaceutical applications. The aim of the project is to elucidate, predominantly by using the technique of two-photon polarization microscopy, various molecular mechanisms of GPCR/G-protein signaling. The project will include developing and improving genetically encoded fluorescent probes of G-protein signaling, using these probes to observe molecular events of G-protein signaling, and obtaining information on relevant molecular mechanisms. Of particular interests will be the ability of one GPCR to signal, under different circumstances, through distinct G-protein pathways (so-called 'biased signaling').
