Topic New electrolyte systems for analysis, purification and characterization of biomolecules and functional organic molecules by capillary and free-flow electromigration methods

Abstract

The aim of the thesis will be to develop new electrolyte systems within a broad pH range, in aqueous and non-aqueous media, and new (pseudo)stationary phases for high-performance analytical and preparative separations of biomolecules (amino acids, peptides, proteins, nucleosides and nucleotides) and functional organic molecules (helicenes and helquats) by capillary and free-flow electromigration methods – zone electrophoresis, isotachophoresis, isoelectric focusing, affinity electrophoresis, electrokinetic chromatography and electrochromatography. A correlation between electromigration in the capillary and in the flow-through chamber will be studied, and based on the correlation model, the analytical capillary electroseparations will be converted into the preparative processes implemented by the above electromigration methods in a free-flow format. In addition to method development for analytical and preparative separations, the further aim will be to develop new methods for physico-chemical and biochemical characterization of biomolecules and functional organic molecules, e.g. for determination of their effective and limiting electrophoretic mobilities, charges, isoelectric points, relative molecular masses, Stokes’ radii, diffusion coefficients, acidity constants of their ionogenic groups and rate constants of their (bio)chemical reactions.
