Leica EM UC7
The Leica EM UC7 ultramicrotome prepares excellent quality semi- and ultra-thin sections, as well as the perfectly smooth surfaces required for light, electron, and atomic force microscopy. The motorized knife stage allows for precise and efficient orientation of the glass or diamond knife via the touchscreen controller. The stereomicroscope observation system features eucentric movement and brightness-controlled LED illumination, allowing precise control of specimen approach and section quality. When attached, the Leica EM FC7 low-temperature sectioning system offers standard, high-gas-flow, and wet sectioning cryo-modes. The anti-vibration system contributes to continual section quality. Up to 100 different specimen and knife profiles can be set.

The Model 1070 Plasma Cleaner applies a low-energy, radio-frequency (RF) plasma that effectively removes hydrocarbon contamination from all materials research specimens. The specimen’s surface is effectively cleaned without changing its elemental composition or structural characteristics. An ultra-clean, oil-free vacuum system ensures optimal specimen cleaning. The Model 1070 readily accepts side-entry specimen holders for all currently available commercial transmission electron microscopes (TEMs), as well as SEM specimens, specimen clamping rings, and other electron microscope accessories. User controllable gas flow, gas mixtures (Argon, Hydrogen, and Oxygen are available), power settings and automated RF matching provide excellent cleaning with very short cycle times.
The Leica EM ACE600 is a high vacuum (≤ 2×10⁻⁶ mbar) coating system that precisely coats samples for subsequent electron microscope (EM) examination. Up to two angled sources can be configured. It is equipped with an automated stage rotation feature, as well as height and tilt adjustment, for optimal material distribution. A quartz crystal measurement accurately determines the layer thickness. The coater offers the convenience of front-door loading and automated, receipt-based coating processes.
The Vitrobot Mark IV offers semi-automated vitrification with two-sided blotting. A controllable ice thickness gradient is achieved through the following available settings: wait time before blotting for sample equilibration on a grid, number of blotting actions (up to 16 times per grid), blotting duration, blot force, longitudinal grid positioning (blot offset), and time between blotting and vitrification (drain time). The working temperature can be set for 4–60° C (at an ambient temperature range between 18–25° C) and is maintained by Peltier controlled heating/cooling. Ultrasonic-controlled humidification enables relative humidity levels of up to 100%.
The Leica EM GP2 Automatic Plunge Freezer offers single sided sensor blotting, required for cell monolayers or support film grids samples. The freezer has fast, easy, and safe filling of the secondary cryogen with the unique liquifying head and controllable secondary cryogen temperature. The environmental chamber provides adjustable temperature (4–60° C) and humidity (up to 99%).
The Leica EM ICE High Pressure Freezer (HPF) freezes samples rapidly (within 20 ms) in liquid nitrogen at high pressure (~ 2000 bar) without ice crystal formation. Samples up to 200 µm in thickness are preserved without artefacts of chemical fixation. The Waffle method sample preparation for cryo-TEM helps to overcome limitations in the size of the sample when using Plunge Freezing. Afterwards, High Pressure Frozen samples can be processed for room temperature electron microscopy (see Leica EM AFS2 with FSP) or analyzed by cryo-TEM or cryo-SEM.
The Leica EM AFS2 is an Automated Freeze Substitution (AFS) and Low Temperature Embedding System for Light and Electron Microscopy. Freeze Substitution (FS) at low temperatures is the follow-on procedure to high pressure freezing (HPF). The Leica EM FSP (Freeze Substitution Processor) is an automatic reagent handling system. Being mounted on the Leica EM AFS2, it automatically dilutes FS media and resins from 100% reagent containers and dispenses the reagents for FS. An integrated LED UV lamp allows immediate polymerization of the sample. Further semi- and ultra-thin sections, as well as the smooth surfaces required for light, electron, and atomic force microscopy can be prepared on Leica EM UC7 ultramicrotome.
